Primers (140–170 bp) for efficient assembly of difficult regions
One-pot digestion & ligation to simplify the workflow and reduce DNA loss
Homologous recombination for seamless, restriction-site-independent cloning
Modular assembly of large genes into ~700–800 bp fragments for greater flexibility
